The protein tyrosine phosphatase 1b as a drug target for the treatment of diabetes type II. Developing effective and selective PTP1B inhibitors
نویسنده
چکیده
PTP1B is a protein tyrosine phosphatase involved in insulin receptor desensitization. PTP1B inhibition, resulting in prolonged maintenance of the activated state of the receptor, practically enhances insulin effect. Thus PTP1B has become a drug target for the treatment of Diabetes type II. Although, a great number of inhibitors have been developed, among which inhibitors binding to the catalytic site and allosteric inhibitors, development of novel highly effective and selective compounds based on profound knowledge of the structural characteristics of the binding sites and inhibitors still remains a challenge.

Protein tyrosine phosphatase 1B (PTP1B) is implicated as a negative regulator of insulin receptor (IR) signaling and a potential drug target for the treatment of type II diabetes and other associated metabolic syndromes. Therefore, small molecular inhibitors of PTP1B can be considered as an attractive approach for the design of new therapeutic agents of type II diabetes diseases. In a continuin...

The Atlas of Diabetes reports 415 million diabetics in the world, a number that has surpassed in half the expected time the twenty year projection. Type 2 diabetes is the most frequent form of the disease; it is characterized by a defect in the secretion of insulin and a resistance in its target organs. In the search for new antidiabetic drugs, one of the principal strategies consists in promot...

Protein tyrosine phosphatase 1B (PTP1B) is a known regulator of the insulin and leptin signaling pathways and is an active target for the design of inhibitors for the treatment of type II diabetes and obesity. Recently, cichoric acid (CHA) and chlorogenic acid (CGA) were predicted by docking methods to be allosteric inhibitors that bind distal to the active site. However, using a combination of...

Diabetes is one of the common metabolic diseases, mainly divided into two types, type 1 diabetes mellitus and type 2 diabetes mellitus. Insulin resistance is the main performance of type 2 diabetes mellitus which are relative to some gene mutation, genetics, obesity and so on. Protein-tyrosine phosphatase 1B (PTP1B) plays an important role as a negative regulator in insulin signaling pathways t...
